Creative Biolabs Develops Versatile Formats of Bispecific Antibodies with Adapted Valency, Structure, Half-life and Biodistribution

Tandem scFvs: Two or more moieties of scFv string together to form tandem scFv with 1+1 (+1…) antigen-binding, fused by a helical linker or a flexible linker. They are extensively used in cancer immunotherapy. BiTEs (bispecific T-cell engagers) are the most famous tandem scFv that comprises an anti-CD3 scFv and an anti-TAA (tumor-associated antigen) scFv to retarget T cells to tumor-associated cells.

Diabody: A dimer of scFv consists of VH and VL domains connected by a short peptide linker. Diabody has monovalent specificity for each antigen (1+1 antigen-binding valency).

Triple Body: It utilizes Fab fragment as scaffold and can lead to 3×1 (trispecific, trivalent), 2+1 (bispecific, bivalent+monovalent), trivalent or 1+1 (bispecific, bivalent) molecule, as well as immunocytokines, immunotoxins, enzyme fusions, etc.

Fab-IgG: Fab-IgG is a format of appended IgGs, the IgG scaffold with Fab fragment in the N terminus and is easy to express owing to less artificial construct and sequence design, which are symmetric bispecific and tetravalent molecules, 2+2 antigen-binding valency.

IgG-scFv: Two scFvs from the same parental antibody are linked to the C terminus of IgG HC. It is bispecifc and tetravalent, 2+2 antigen-binding valency and can be custom-tailored to meet customers’ specific requirements.

TandAb: TandAb is expressed as a monomeric subunit (single chain Diabody, scDb) with four variable domains from two parental antibodies.

Single-chain Diabody (scDb): It is a derivative of Diabody that connects the first chain and the second chain of diabodies by a flexible linker, transforming the heterodimeric format of diabody into a single-chain format. scDb can also be fused with Fc or CH3 domain to exert Fc-mediated functions and/or extend half-life.

Single-chain triplebody (Sctb): It has three tandem scFv units, bispecific or trispecific, 2+1 or 1+1+1 antigen-binding valency and allows the recruitment of immune cells to eliminate disease cells, such as tumor cells.

Tandem Fabs: It comprises two Fabs targeting different epitopes and is bispecific and has monovalent for each antigen specificity (1+1 antigen-binding valency).

CrossMab: It remains the general structure and size of a conventional IgG. In CrossMAb, KIH (knobs-into-holes) technology is used to permit the heavy chain heterodimerize. Usually, the referred CrossMab is a bispecific, bivalent molecule, 1+1 antigen-binding valency.
