Cell lysis is the first step in cell isolation, organelle isolation and protein extraction and purification. Therefore, cell lysis has opened the door to countless proteomic research methods. Physical lysis was the preferred method of cell disruption and cell content extraction in the history, however, it often required expensive and cumbersome equipment and involved difficult-to-repeat protocols due to the variability of the equipment. In addition, the traditional methods of physical destruction are not conducive to the high throughput and small volume typical of modern laboratory research.

A number of cell lysis methods have been developed to obtain different types of organisms, sample types (cells or tissues), and optimal yield and purity of target molecules or subcellular structures. Cell lysis methods can be divided into two broad categories, namely reagent-based and physical destruction. In recent years, detergent-based cracking methods have been commonly used, different detergent-based solutions have been developed that consist of specific types and concentrations of detergents, buffers, salts, and reducing agents to provide the best possible results for a particular species and cell type through experimental and erroneous experimental testing.

Cell lysis interferes with the cellular environment and destabilizes endogenous proteases and phosphatases so that the extracted proteins are degraded or artificially modified by the activity of these molecules. In order to prevent these effects and obtain the best possible protein yield in cell lysis, protease and phosphatase inhibitors were added to the lysis reagent. Many compounds have been identified and applied to inactivate or block the activity of proteases and phosphatases by binding them reversibly or irreversibly.

In Creative Biogene, cell lines were designed as double knockout lines through CRISPR technology, and double knockout genotypes were verified with PCR followed by sequencing. Knockout cell lysates were cell homogenates in RIPA buffer prepared from the KO cell line. A bottle of lysate from the parental cell line was also provided as an internal control. Besides, Creative Biogene has a large library of lysates such as HEK293, HEK293T, HeLa, insect cells, etc., which were generated by transfection/transduction of foreign protein coding sequences into these cells, and then homogenized using RIPA lysis buffer. These cell lysates can be used as effective positive controls for the verification of newly generated antibodies against antigens such as humans and viruses.
